T-cell immunology is at the core of infectious diseases, cancer, and allergy. We have projects concerning herpes simplex viruses types 1 and 2, varicella zoster virus, vaccinia (the vaccine for smallpox), Merkel cell polyoma virus, and HHV-6, and a project on T cell-mediated severe drug reactions. These pathogens mostly have large genomes, so determining T-cell antigens is challenging. Our technical expertise is various molecular methods to interrogate T-cell responses to a very high level of definition. A suite of modern immunology tools such as intracellular cytokine cytometry, tetramers, T cell cloning, artificial APC, cell killing assays, transgenic TCR expression, etc. are in use to correlate immune variables with outcomes or immune responses to vaccination. HSV-1 and HSV-2 vaccine candidates have been discovered, studied in mice, and entered human trials. We perform immune monitoring for human clinical trials of candidate HSV vaccines. For Mycobacterium tuberculosis (MTB), I am responsible for the IGRA testing at our medical center, and have mentored several trainees interested in this T-cell immunology clinical test.
